Extracellular vesicles (EVs), laden with biomolecules from their originating cells, present a promising novel source of disease biomarkers, and their clinical applications are being intensely investigated. Tunable resistive pulse sensing (TRPS) uses the magnitude of a small ionic resistive pulse current to ascertain the dimensions, concentration, and zeta potential of EVs. Commonly, environmental noise serves as a limiting factor in the accuracy of measurements made by sensing devices. TRPS’s vulnerability to environmental noise, which includes both mechanical and electrical sources, is pronounced. The maximum detectable amount by TRPS is determined by the physical size of its elastomeric tunable nanopore. The electrical signal-to-noise ratio dictates the lowest possible limit. Employing simulation techniques, we developed an external instrument to dampen environmental sounds for precise TRPS measurements. Subsequent to the shield’s use, a decrease in environmental noise, including mechanical and electrical sources, was observed. The study’s findings confirmed the shield’s noise reduction, ascertained by measuring EVs from different cell types. By deploying the shield, the detection of extracellular vesicles (EVs) measuring less than 200 nanometers was markedly enhanced, thereby addressing the difficulties encountered by conventional methods of quantification. Myotonic dystrophy type 1 (DM1), a neuromuscular disorder, is directly linked to the expansion of CTG microsatellites within the 3′ untranslated region of the DMPK gene, triggering the expression of transcripts harboring expanded CUG repeats (CUGexp). A toxic RNA gain of function, characterized by the formation of nuclear CUGexp RNA aggregates, explains the pathophysiology. These aggregates sequester and alter MBNL splicing factors, causing splicing misregulation and the subsequent DM1 symptoms. In the case of DM1, a cure is yet to be discovered, and most treatment strategies center on removing CUGexp-DMPK transcripts. This study investigates a DMPK-promoter silencing strategy, using CRISPR interference as a novel alternative method. The examination of different sgRNAs that target the DMPK promoter happens inside the muscle cells of individuals with DM1. Although microRNAs (miRNAs) are well-known regulators of cellular processes, their specific impact on Leydig cell (LC) activity remains inadequately understood. We detected a considerable difference in the levels of miR-300-3p expression during the differentiation process of progenitor Langerhans cells (PLCs) into adult Langerhans cells (ALCs). PLC proliferation was enhanced and testosterone production was decreased by high miR-300-3p expression in PLCs, achieved through the specific targeting of the steroidogenic factor-1 (Sf-1) and transcription factor forkhead box O1 (FoxO1) genes, respectively. The differentiation of PLCs into ALCs corresponded with a substantial reduction in miR-300-3p levels, thereby stimulating testosterone synthesis and inhibiting ALC proliferation through the upregulation of SF-1 and FoxO1 expression. The miR-300-3p expression was regulated by the LH/METTL3/SMURF2/SMAD2 cascade pathway, where luteinizing hormone (LH) amplified SMAD-specific E3 ubiquitin protein ligase 2 (SMURF2) expression via METTL3-catalyzed N6-methyladenosine modification of Smurf2. By obstructing the attachment of SMAD2 to the miR-300 promoter, Smurf2 consequently limited the production of miR-300. Of note, a relationship existed between miR-300-3p and obesity-related testosterone deficiency in men, and the blocking of miR-300-3p effectively prevented the decline in testosterone levels in obese mice. The data suggests that miR-300-3p plays a key role in both the differentiation and the function of LC cells, and holds potential as a diagnostic or therapeutic target in addressing testosterone deficiency linked to obesity.

Transcription of the Mfrp and C1-tumor necrosis factor related protein 5 (Ctrp5) genes, as a bicistronic unit, is linked to retinal degeneration in retinal pigment epithelium (RPE) cells when either gene is dysregulated. Yet, the mechanisms by which the expression of the bicistronic message is controlled remain in question. Employing microRNA analysis, we detected a negative feedback loop that controls the expression level of the bicistronic Mfrp and Ctrp5 transcript to ensure normal levels. More specifically, the microRNA miR-149-3p, a conserved molecule, attaches itself to the 3′ untranslated region of the Mfrp gene. In rd6 mice lacking MFRP, the expression of miR-149-3p was lower than in wild-type mice, contributing to an increase in the bicistronic transcript’s abundance. This report details a capsid-modified rAAVDJ-3M vector, successfully transducing RPE cells in a powerful and specific manner after subretinal injection. The parental vector and modified vector showed similar serum anti-rAAV antibody levels, yet the modified vector exhibited a decreased number of microglial infiltrations. Our results compellingly indicate that a strategy combining MFRP overexpression with the knockdown of the bicistronic transcript yielded more significant vision recovery than MFRP overexpression alone. Social and therapeutic ramifications of chronic wounds remain a key obstacle in their resolution, necessitating comprehensive and sustained medical attention. Recent theragnostic efforts, though focused on wound management, do not adequately facilitate the healing of chronic wounds. A marked increase in cases of under- or over-healing wounds calls into question the efficacy of current wound management, demanding an investigation into alternative treatment methods to address this significant unmet need. To compensate for this gap in wound treatment, extracellular vesicles (EVs), designed for targeted delivery of therapeutic agents, are emerging as a promising treatment option for both acute and persistent injuries. To address these challenges, we analyze the foundational biology of EVs, explore their associated pharmacology, interpret immunogenic consequences, and investigate the promise of enhanced, long-term wound healing strategies, thoroughly considering their potentially unacceptable side effects. The therapeutic function of EVs in severe wound infections, modulated through biological regulation and in combination with biomaterials and drug carriers, opens avenues for the development of novel therapies for this long-term condition, with a particular focus on biomaterial-based drug delivery systems. Clinical translation of EV-based wound management treatments, with its accompanying hurdles and potential outcomes, suggests a possible future therapeutic application.
