A recent study reported that MKRN3 interacts with and suppresses neural pentraxin-1 precursor (NPTX1) activity via polyubiquitination during early puberty in the mouse hypothalamus. Objective This study investigated the correlation between serum NPTX1 and MKRN3 in CPP girls and predicted the potential role of NPTX1 in pubertal progression. Methods In this case-control study, we examined 34 girls diagnosed with CPP and 34 healthy prepubertal girls. Anthropometric and hormonal parameters were measured and serum levels of NPTX1 and MKRN3 were evaluated with commercial enzyme-linked immunosorbent assay kits. Results Serum MKRN3 level decreased significantly in CPP patients compared to controls (344.48 ± 333.77 and 1295.21 ± 780.80 pg/mL, respectively, p less then 0.001). Serum MKRN3 tended to decrease as Tanner breast stage increased. However, no significant difference was observed in serum NPTX1 levels between patients and controls (20.14 ± 31.75 ng/mL and 12.93 ± 8.28 ng/mL, respectively, p=0.248). The serum level of NPTX1 did not change significantly with the Tanner breast stage. Serum NPTX1 was correlated with the height standard deviation score (r=0.255; p less then 0.05), but was not correlated with serum MKRN3 level or the others. Conclusion Although serum NPTX1 level was independent of serum MKRN3 level, the possibility they might be involved in the progression of puberty or CPP remains. Although the obtained sequences were genetically different from each other, it was found that genetic variations were limited.This study aims to analyze and assess the effects of three particular dentin pretreatment solutions on the bond durability of a two-step self-etch adhesive (Optibond XTR) applied to dentin after ageing for 2 years. Thirty-five third molars which were extracted (n = 5 for μTBS, n = 2 for nanoleakage) were divided into five groups Group 1Control (no pretreatment), Group 217% EDTA, Group 32% CHX, Group 417% EDTA plus 2% CHX, Group 5 Q-Mix. After the pretreatments of dentin, the dentin adhesive was applied as per the guidelines provided by the manufacturer. Half of the specimens were subjected to μTBS tests for 24 hr, while the remaining half were subjected to the tests after being kept for 2 years in water storage. Also, nanoleakage was evaluated with FE-SEM by examining silver nitrate deposits. The data obtained were evaluated using a two-way analysis of variance and Tukey Post Hoc test. The dentin pretreatments did not affect the 24 hr and 2 years μTBS values for OptiBond XTR. At 24 hr, the EDTA + CHX group (50.3 ± 4.9) showed that the highest μTBS value was obtained. Water ageing significantly reduced the μTBS results and after 2 years the highest μTBS value was obtained from the Q-Mix group (37.7 ± 5.2). Different dentin pretreatments do not alter the 24-hr μTBS and were not able to preserve the bond strength after 2 years of ageing. Q-Mix was able to slow down the regression in the strength of the dentin bond as well as nanoleakage over time.Adolescent idiopathic scoliosis (AIS), a sideways curvature of the spine, is the most common pediatric musculoskeletal disorder, affecting ~ 3% of the population worldwide. However, its genetic bases and tissues of origin remain largely unknown. Several genome-wide association studies (GWAS) have implicated nucleotide variants in noncoding sequences that control genes with important roles in cartilage, muscle, bone, connective tissue and intervertebral discs (IVDs) as drivers of AIS susceptibility. Here, we set out to define the expression of AIS-associated genes and active regulatory elements by performing RNA-seq and ChIP-seq against H3K27ac in these tissues in mouse and human. Our study highlights genetic pathways involving AIS-associated loci that regulate chondrogenesis, IVD development and connective tissue maintenance and homeostasis. In addition, we identify thousands of putative AIS-associated regulatory elements which may orchestrate tissue-specific expression in musculoskeletal tissues of the spine. Quantification of enhancer activity of several candidate regulatory elements from our study identifies three functional enhancers carrying AIS-associated GWAS SNPs at the ADGRG6 and BNC2 loci. Our findings provide a novel genome-wide catalog of AIS-relevant genes and regulatory elements and aid in the identification of novel targets for AIS causality and treatment.The solvent effect usually plays an important role in triplet-triplet annihilation (TTA) upconversion processes. In this work, we have studied the TTA upconversion kinetics of perylene with Bodipy-phenyl-C60 as the triplet photosensitizer in five solvents, 1,4-dioxane, dichlorobenzene, chlorobenzene, toluene, and tetrahydrofuran (THF). Although no significant solvent effect was observed in steady-state absorption and fluorescence emission spectra, the overall TTA upconversion quantum yields showed a profound dependence on solvent properties, i.e. 4.9% in 1,4-dioxane, 7.1% in dichlorobenzene, 6.7% in chlorobenzene, 4.6% in toluene, and 2.2% in THF (the maximum of 50%). Each elementary reaction step involved in the overall process was analyzed by applying femtosecond and nanosecond time-resolved transient absorption spectroscopy, revealing that the fluorescence emission of perylene was more significantly affected by the solvents in contrast to the other steps. Moreover, an extra intramolecular energy-transfer pathway of Bodipy-phenyl-C60 was found via the formation of charge-separated states in dichlorobenzene, chlorobenzene, and THF solvents, once being excited.
